{"id":2389,"date":"2020-09-21T13:19:22","date_gmt":"2020-09-21T11:19:22","guid":{"rendered":"https:\/\/meddists.com\/learn\/pre-clinical\/biochemistry\/cell-proliferation-and-apoptosis\/the-restriction-point-control-and-the-role-of-p53-in-the-life-of-cells\/"},"modified":"2020-09-21T13:22:18","modified_gmt":"2020-09-21T11:22:18","slug":"the-restriction-point-control-and-the-role-of-p53-in-the-life-of-cells","status":"publish","type":"page","link":"https:\/\/meddists.com\/learn\/pre-clinical\/biochemistry\/biochemistry-of-the-tissues\/cell-proliferation-and-apoptosis\/the-restriction-point-control-and-the-role-of-p53-in-the-life-of-cells\/","title":{"rendered":"The restriction point control and the role of p53 in the life of cells"},"content":{"rendered":"\n<ul class=\"wp-block-list\"><li>The DNA damage checkpoint is extremely important because it can hold the cell in G1 or induce apoptosis or if the size of the defect is manageable, to correct the defect.<\/li><li>The ATM protein transmits information about DNA damage to p53, which induces the CDK inhibitor p21, which holds the cell in G1.<\/li><li>P53 contains separate domains for transcription activation, DNA binding, oligomerization, and binding to damaged DNA.<\/li><li>MDM2 binding to p53 inhibits transactivation and degrades p53 \u2013 however, by ATM, DNA-PK, ATR serine phosphorylation, they can inhibit MDM2 binding, stabilizing p53.<\/li><\/ul>\n\n\n\n<p class=\"wp-block-paragraph\"><strong>P53 is activated by several effects:<\/strong><\/p>\n\n\n\n<ol class=\"wp-block-list\"><li>DNA damage<\/li><li>hypoxia<\/li><li>abnormal proliferative signals<\/li><li>DNA replication errors<\/li><\/ol>\n\n\n\n<ul class=\"wp-block-list\"><li>When DNA is exposed to damaging agents and UV radiation, it is stabilized by N-terminal serine phosphorylation and activates its DNA binding capacity (the C-terminal portion is acetylated and dephosphorylated).<\/li><li>Hypoxia and hypoglycemia both lead to the stabilization of p53 by phosphorylation, while inhibition of oncogenic stimuli by MDM2.<\/li><\/ul>\n\n\n\n<p class=\"wp-block-paragraph\"><strong>Plenty of genes regulated by p53 TF (DNA repair, cell cycle arrest, cell death killer \u2013 50% of p53 in cancer)<\/strong><\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Its DNA binding domain binds to specific sequences and initiates transcription of substances:<\/p>\n\n\n\n<ol class=\"wp-block-list\"><li>CDK inhibitors<\/li><li>error repair enzymes<\/li><li>miR-34<\/li><li>interacts with Bcl-2, an inhibitor of apoptosis<\/li><li>activates the protein complex formed by miRNAs that inhibit cell proliferation<\/li><li>if it enters the nucleus and initiates cell death, then among the genes responsible for apoptosis, BAX, PUMA, NOXA will be activated<\/li><\/ol>\n\n\n\n<div class=\"wp-block-image\"><figure class=\"aligncenter\"><a href=\"https:\/\/meddists.com\/wp-content\/uploads\/2020\/03\/p53.jpg\" target=\"_blank\" title=\"The restriction point control and the role of p53 in the life of cells\"><img decoding=\"async\" src=\"https:\/\/meddists.com\/wp-content\/uploads\/2020\/03\/p53.jpg\" alt=\"\" class=\"wp-image-26019\"\/><\/a><\/figure><\/div>\n\n\n\n<p class=\"wp-block-paragraph\"><strong>Figure 1. p53 \u2013 Copyright \u00a9 Meddists.com<\/strong><\/p>\n<div id=\"the_titles\" style=\"display:none;\"><\/div>","protected":false},"excerpt":{"rendered":"<p>The DNA damage checkpoint is extremely important because it can hold the cell in G1 or induce apoptosis or if the size of the defect is manageable, to correct the defect.
